## What it is

Urolithin A is a natural compound produced by gut bacteria from ellagitannins and ellagic acid, dietary polyphenols found in pomegranates, walnuts, and berries. It is not present directly in food but is generated during digestion, and the capacity to produce it varies widely between individuals depending on gut microbiome composition. Mechanistically, urolithin A stimulates mitophagy (the cellular recycling of damaged mitochondria) and promotes mitochondrial biogenesis and function.

## Its role in longevity

In human research, urolithin A supplementation is safe and bioavailable and improves markers of mitochondrial health. Randomized trials report better muscle endurance in adults 65-90, roughly 12% strength gains with improved aerobic endurance in middle-aged adults, lower inflammatory markers (C-reactive protein, acylcarnitines), and, after just four weeks in a 2025 trial, a shift toward younger immune profiles with expanded naive-like, less-exhausted CD8+ T cells. Honest caveats: nearly all trials are small (about 50-100 people), short (up to 4 months), and manufacturer-funded; several primary endpoints were missed (peak power, maximal ATP production, and, in one trial, 6-minute walk distance); and no long-term disease or mortality data exist. This evidence describes the urolithin A compound and its mechanism rather than any specific commercial product.

- **Dose used in studies:** 1,000 mg/day in most trials (500–1,000 mg/day studied)
- **What this grade does not say:** Several small, well-controlled randomized trials show improved muscle endurance and strength, lower inflammatory markers, and younger-looking immune-cell profiles, with an excellent safety record. But nearly all are short (4 months or less) and manufacturer-funded, some missed their primary endpoints, and there is no long-term disease or lifespan data. Most people cannot reliably make urolithin A from food — the rationale for direct supplementation.
