## What it is

Sulforaphane is an isothiocyanate phytochemical formed when the plant enzyme myrosinase acts on glucoraphanin, a glucosinolate concentrated in cruciferous vegetables such as broccoli and especially young broccoli sprouts. In the body it is a potent activator of the NRF2 (NF-E2-related factor 2) transcription factor, which induces phase-2 antioxidant and detoxification enzymes. In diabetes research it has been shown to suppress hepatic glucose production by promoting NRF2 nuclear translocation and downregulating key gluconeogenic enzymes. It is commonly delivered as a standardized broccoli sprout extract.

## Its role in longevity

Human trials indicate that concentrated broccoli-sprout sulforaphane can modestly lower fasting blood glucose and glycated hemoglobin, most clearly in obese or dysregulated type 2 diabetes and in a subset of people with prediabetes; a 2025 randomized placebo-controlled trial found a small average reduction in fasting glucose (about 0.2 mmol/L), with substantially larger effects in participants whose gut microbiota and metabolic profile predicted response. A separate line of randomized trials shows sulforaphane-rich preparations enhance the detoxification and excretion of airborne and tobacco-derived carcinogens, and early trials report improved insulin resistance and liver enzymes, with a generally good safety and tolerability profile aside from mild gastrointestinal effects. Benefits so far are biomarker-level rather than proven disease-outcome reductions, and supplement bioavailability varies widely unless the preparation retains active myrosinase. This evidence concerns the sulforaphane compound and broccoli-sprout extracts in general, not this specific commercial product.

## Evidence grade: Moderate evidence

- **What the grade means:** Curated grade based on the human and mechanistic evidence for this compound.
- **What this grade does not say:** Randomized trials show real biomarker effects — enhanced carcinogen detoxification and a modest glucose-lowering signal in prediabetes — but these are surrogate markers, not proven disease-outcome reductions. Actual sulforaphane delivery varies widely between supplements unless active myrosinase is retained. Mild gastrointestinal upset is the main side effect.
