## What it is

Curcumin is the principal curcuminoid and yellow pigment of turmeric (Curcuma longa), a rhizome long used as a culinary spice and in traditional medicine. It is a polyphenol studied for anti-inflammatory and antioxidant activity, acting on signaling pathways such as nuclear factor-kappaB and lowering circulating inflammatory mediators including C-reactive protein (CRP), interleukin-6 (IL-6) and tumor necrosis factor-alpha (TNF-alpha). Because native curcumin is poorly water-soluble, rapidly metabolized and poorly absorbed, 'high-bioavailability' products use formulations such as piperine co-administration, nanoparticles, or lipid/phospholipid carriers to raise plasma levels.

## Its role in longevity

Meta-analyses of randomized controlled trials generally find that curcumin/turmeric supplementation lowers CRP, IL-6 and TNF-alpha and improves oxidative-stress markers, with symptomatic benefits reported in osteoarthritis and rheumatoid arthritis, modest LDL-cholesterol and triglyceride reductions in cardiometabolic populations, and adjunctive antidepressant effects; one small 18-month trial of a bioavailable formulation reported improved verbal memory and reduced brain amyloid/tau signal on PET imaging in non-demented adults. However, effects on individual cytokines (for example IL-6 or CRP in some populations) are inconsistent, several analyses report null results, and trials are often small, short, and heterogeneous in formulation. Higher-bioavailability formulations such as nanocurcumin appear to enhance some effects, but the same absorption-boosting property (often piperine, which inhibits hepatic metabolism) has been linked to a growing case series of immune-mediated liver injury, so prolonged use warrants caution. This evidence describes the curcumin compound and its formulations as studied in the literature, not this specific commercial product.
