## What it is

Pomegranate ellagitannin extract is a concentrated source of ellagitannins (such as punicalagins) and ellagic acid, the polyphenols responsible for much of the pomegranate's antioxidant activity. After ingestion, these compounds are hydrolyzed to ellagic acid and then metabolized by the gut microbiota into urolithins, chiefly urolithin A. Urolithin A is a postbiotic that stimulates mitophagy, the cellular quality-control process that recycles damaged mitochondria, and the capacity to produce it varies substantially between individuals depending on gut microbiome composition.

## Its role in longevity

In human research, the ellagitannin-derived metabolite urolithin A has improved muscle strength, endurance, and biomarkers of mitochondrial function and inflammation in randomized trials of middle-aged and older adults, and a small 2025 trial found more youthful immune-cell profiles after four weeks, although several primary endpoints (e.g., peak power, maximal ATP production) were not met. Separately, pomegranate juice and extracts have modestly lowered blood pressure (a few mmHg on average) in meta-analyses of randomized trials — though at least one systematic review rates that evidence as weak — and a year-long trial found daily juice helped preserve visual memory while placebo declined. The mitophagy findings come mainly from the purified metabolite rather than the extract itself, and urolithin production varies widely between individuals. This evidence describes the ellagitannins, urolithin A, and the pomegranate polyphenols themselves, not this specific commercial extract product.

## Evidence grade: Moderate evidence

- **What the grade means:** Curated grade based on the human and mechanistic evidence for this compound.
- **What this grade does not say:** Human trials and meta-analyses support small blood-pressure reductions (a few mmHg) and one year-long trial suggests memory preservation, but at least one systematic review calls the blood-pressure evidence weak. The headline mitophagy benefits were shown mainly with purified urolithin A, not pomegranate extract, and microbiome conversion of ellagitannins varies widely — many people are low producers.

## Where it comes from without a bottle

Food, exercise and behaviour routes to the same pathway, each with its citation.

| Natural source | How it works | Citation |
| --- | --- | --- |
| Pomegranate and walnuts (ellagitannin foods) | Your gut bacteria convert the ellagitannins in pomegranate and walnuts into urolithins, the metabolites behind the benefits, so eating these whole foods is the natural precursor route. | Molecular Nutrition & Food Research 2022 [PMID 35118817](https://pubmed.ncbi.nlm.nih.gov/35118817/) |
| Walnuts or pomegranate in human trials | In people, eating walnuts or taking pomegranate extract measurably produced urolithins in urine, confirming these everyday foods actually deliver the active metabolite, though output varies by gut metabotype. | Clinical Nutrition 2017 [PMID 28347564](https://pubmed.ncbi.nlm.nih.gov/28347564/) |
