## What it is

Grape seed extract (GSE) is a polyphenol-rich preparation made from the seeds of wine grapes (Vitis vinifera), standardized primarily for proanthocyanidins (oligomeric procyanidins) along with catechins and other flavonoids. Marketed as a dietary supplement, it has antioxidant and anti-inflammatory activity. Preclinical work indicates that its proanthocyanidins can increase endothelial nitric oxide synthase (eNOS) expression and nitric oxide production, in part through AMPK/SIRT1/KLF2 signaling, and can lower endothelin-1 and oxidative stress, which together support endothelial function and vascular tone.

## Its role in longevity

Randomized trials and meta-analyses indicate that grape seed extract can modestly lower blood pressure — pooled systolic reductions of roughly 2–6 mmHg, larger in younger, obese, or metabolic-syndrome patients — and may improve markers of blood glucose, LDL cholesterol, triglycerides, and inflammation. Two small 2024 trials in non-alcoholic fatty liver disease reported improved insulin resistance, liver enzymes, oxidative stress, and quality of life, though both were single-center studies of about 50 people. Effects on flow-mediated dilation are inconsistent, with some trials and one large meta-analysis showing no significant change, and a 12-week trial in healthy young adults found no cognitive benefit. The literature is limited by small, heterogeneous, often short and sometimes industry-funded studies, and there is no evidence on lifespan or hard cardiovascular outcomes. This evidence characterizes the grape seed extract compound and its active proanthocyanidins in general, not this specific commercial product.
