## What it is

NOVOS Core is a multi-ingredient dietary supplement marketed for longevity that combines about a dozen compounds intended to target several biological 'hallmarks of aging.' Its ingredients include calcium alpha-ketoglutarate (a Krebs-cycle metabolite), the senolytic flavonoid fisetin, pterostilbene (a methylated resveratrol analog), glycine, glucosamine sulfate, magnesium malate, L-theanine, Rhodiola rosea, a microdose of lithium, hyaluronic acid, ginger, and vitamin C. Rather than acting through a single pathway, the formula is designed so that different components address distinct aging processes such as cellular senescence, mitochondrial and metabolic function, epigenetic regulation, and oxidative stress. The evidence below concerns these individual active ingredients and their mechanisms, since the finished multi-ingredient product has not itself been tested in controlled human trials.

## Its role in longevity

Several of NOVOS Core's ingredients have genuine, though mostly early-stage, human or animal evidence relevant to aging: trials of GlyNAC (glycine plus N-acetylcysteine) improved oxidative stress, mitochondrial function, strength and walking performance in older adults — though at glycine doses far above what the sachets provide and paired with NAC, which the formula lacks; a small retrospective analysis linked an alpha-ketoglutarate formulation to reduced DNA-methylation age, with a proper randomized trial still underway; regular glucosamine use was associated with about 15% lower all-cause mortality in the 495,000-person UK Biobank cohort, an observational finding that cannot prove cause; low-dose lithium correlated with lower all-cause mortality in a large population; and fisetin extended lifespan and reduced senescent-cell burden in mice, with human senolytic trials still in progress. Magnesium modestly lowers blood pressure in pooled trials, but pterostilbene is a weak spot: it raised LDL cholesterol by about 17 mg/dL in one randomized trial, and pterostilbene combined with nicotinamide riboside did not improve muscle recovery in elderly volunteers. The specific doses, combination, and long-term safety of this exact stack have not been validated. This evidence describes the individual compounds and their mechanisms, not this specific commercial multi-ingredient product, which has not been tested as a whole in controlled human studies.
