## What it is

Zinc picolinate is a chelated form of the essential trace mineral zinc, in which zinc is bound to picolinic acid and sold as an oral dietary supplement. Zinc is a catalytic and structural cofactor for hundreds of enzymes and transcription factors and is required for normal immune-cell development and function, wound healing, and retinal antioxidant defense. In the setting of the common cold, ionic zinc is thought to interfere with rhinovirus replication and to modulate inflammatory and immune responses in the upper respiratory tract, and adequate zinc status matters more with advancing age, when deficiency becomes common. Picolinate is marketed for superior absorption, but no head-to-head human trials establish it over other zinc forms, so the evidence summarized here applies to zinc generally.

## Its role in longevity

Multiple meta-analyses of randomized trials indicate that oral zinc, started soon after symptom onset, can modestly shorten the duration of common-cold symptoms, although a 2024 Cochrane update rated the certainty of this evidence low and the effect remains genuinely contested. Separately, in the large AREDS randomized trial, high-dose zinc combined with antioxidant vitamins slowed progression to advanced age-related macular degeneration in at-risk older adults, though the follow-up AREDS2 trial found no advantage to altering the zinc dose. Bad taste and nausea are frequent with lozenges, chronic high doses can cause copper deficiency, a preventive benefit is not established, and immune or longevity benefits in zinc-replete adults are weakly supported. This evidence concerns zinc and its compounds in general, not this specific commercial zinc picolinate product.

Otherwise strong trial evidence exists only for the AREDS eye formulation; immune or longevity benefits in zinc-replete adults are weakly supported, and no trials show the picolinate form is superior.
