## What it is

GlyNAC is a nutritional supplement combining two amino acids, glycine and N-acetylcysteine (NAC, a precursor of cysteine), that together supply the building blocks the body needs to synthesize glutathione, its most abundant intracellular antioxidant. Glutathione levels and synthesis decline with age, and this deficiency has been linked to elevated oxidative stress and mitochondrial dysfunction, two proposed drivers of aging. By providing both precursor amino acids, GlyNAC is intended to restore glutathione production and, through it, counter oxidative stress and support mitochondrial function. Most of the human research on GlyNAC has been conducted by a single group at Baylor College of Medicine.

## Its role in longevity

In small clinical trials, mostly pilot and open-label studies plus one randomized placebo-controlled trial of 24 older adults from the same laboratory, GlyNAC supplementation corrected glutathione deficiency and improved markers of oxidative stress, mitochondrial function, inflammation, insulin resistance, muscle strength, gait speed and cognition, and in mice it extended lifespan by about 24%. However, the human evidence base is limited by small samples, short duration and reliance on one research group, and an independent randomized dose-ranging trial in 114 healthy older adults found glutathione and oxidative-damage benefits mainly in participants with elevated baseline oxidative stress, so effects may be context-dependent; measured benefits also reversed after supplementation stopped. This evidence describes the glycine/NAC/glutathione mechanism and the GlyNAC combination studied in research settings, not this specific commercial product.

## Evidence grade: Emerging evidence

- **What the grade means:** Curated grade based on the human and mechanistic evidence for this compound.
- **Dose used in studies:** In the Baylor/Sekhar trials, older adults took glycine 1.33 mmol/kg and N-acetylcysteine 0.83 mmol/kg daily (~100 mg/kg of each, roughly 7 g + 7 g for a 70 kg adult) for 16 weeks.
- **Dose from food:** Glutathione itself isn't absorbed intact from food; the body builds it from precursors. Glycine is plentiful in collagen, meat and gelatin, but NAC is synthetic and not a food component, so trial-level intake is hard to reach by diet alone.
- **What this grade does not say:** Small human trials show GlyNAC restores glutathione and improves oxidative-stress, mitochondrial and physical-function markers, but nearly all come from one research group, the largest placebo-controlled trial enrolled 24 older adults for 16 weeks, and benefits faded after stopping. The ~24% lifespan extension was shown only in mice; there are no human data on disease or mortality outcomes.
