## What it is

Glutathione (GSH) is a tripeptide of glutamate, cysteine, and glycine synthesized inside virtually all human cells, where it serves as the body's most abundant intracellular antioxidant. It neutralizes reactive oxygen species, supports detoxification and immune function, and maintains cellular redox balance, cycling between reduced (GSH) and oxidized (GSSG) forms. Intracellular glutathione concentrations tend to fall with age and in states of high oxidative stress, which has prompted interest in raising glutathione through direct oral, sublingual, or liposomal supplements, or through precursor amino acids such as N-acetylcysteine and glycine.

## Its role in longevity

Randomized trials show that oral glutathione supplementation can genuinely raise body stores — a well-run 6-month trial found dose-dependent increases of about 30–35% in blood cells at 1,000 mg/day, along with a more than twofold rise in NK-cell activity — and lower oxidative-stress markers, answering historical doubts about oral bioavailability, though results vary by formulation and the supporting liposomal pilot was small and uncontrolled. Supplying precursors as GlyNAC (glycine plus N-acetylcysteine) improved glutathione levels, mitochondrial function and several aging hallmarks in older adults in one small 16-week trial, but at least one controlled trial found no overall rise in glutathione except in participants with high oxidative stress. For skin, a 4-week trial found 500 mg/day lowered melanin index modestly — significantly at only two of six sites — and reviews rate the lightening evidence as weak, with long-term and IV safety unestablished. Researchers emphasize that taking glutathione is not the same as supplying its precursors, and no trial shows disease or longevity outcomes. This evidence describes glutathione and its precursor biology in general, not this specific commercial product.

- **What the grade means:** Curated grade based on the human and mechanistic evidence for this compound.
- **Dose used in studies:** Liposomal glutathione trials used 500–1,000 mg/day (Sinha 2018) and 300 mg in a 2026 crossover; by contrast, plain oral glutathione up to 3,000 mg barely raised blood levels (Witschi 1992) because gut enzymes degrade it.
- **Dose from food:** Small amounts occur in fresh produce, raw meat and fish, but dietary glutathione is largely broken down by gut peptidases before absorption, so food isn't a reliable way to raise body glutathione versus precursor or liposomal approaches.
- **What this grade does not say:** Oral glutathione genuinely raises body stores and lowers oxidative-stress markers in randomized trials (about 30–35% at 1,000 mg/day over 6 months), with early immune-marker signals. Clinical outcomes are thin: skin-lightening effects are modest and inconsistent, and no trial shows disease or longevity benefit.
