## What it is

Methylsulfonylmethane (MSM), also called dimethyl sulfone or methyl sulfone, is a naturally occurring organosulfur compound found in small amounts in fresh vegetables, fruits, grains, and animal products, and is also manufactured synthetically for use as a dietary supplement. It provides a source of bioavailable sulfur and is the oxidized metabolite of dimethyl sulfoxide (DMSO). Laboratory and animal research indicates anti-inflammatory and antioxidant activity, including inhibition of NF-κB signaling and NLRP3 inflammasome activation and support of glutathione and total antioxidant capacity. It is most often taken orally, typically at about 1.5-6 g per day, for joint and muscle health.

## Its role in longevity

Across human studies, MSM has been evaluated mainly for osteoarthritis, exercise recovery, and seasonal allergic rhinitis. Multiple randomized, placebo-controlled trials and meta-analyses report statistically significant but generally small reductions in joint pain and improvements in physical function - alone or combined with glucosamine, chondroitin, or boswellic acids - while small trials suggest it may attenuate post-exercise muscle and joint pain, oxidative stress and inflammatory signaling; it is a GRAS (Generally Recognized As Safe) substance well tolerated at doses up to about 4 g per day. The evidence is limited by small samples, short duration, and inconsistent results, with several trials and evidence reviews reporting null findings or effects of uncertain clinical importance; effects appear smaller and slower than NSAIDs, there is no evidence of disease modification, and no confirmed long-term benefit - 'supports joint comfort' is a fairer summary than 'rebuilds cartilage'. This evidence describes the compound methylsulfonylmethane and its mechanisms rather than this specific commercial product.

## Evidence grade: Moderate evidence

- **What the grade means:** Curated grade based on the human and mechanistic evidence for this compound.
- **Dose used in studies:** 2-6 g/day (typical studied range)
- **What this grade does not say:** Small randomized trials show statistically significant but clinically mild improvements in knee pain and function over 8-12 weeks, with some null results. There is no evidence of disease modification or anti-aging benefit, and effects look smaller and slower than NSAIDs. Safety is excellent (GRAS) at typical doses.

## Where it comes from without a bottle

Food, exercise and behaviour routes to the same pathway, each with its citation.

| Natural source | How it works | Citation |
| --- | --- | --- |
| Sulfur-rich whole foods (garlic, onion, eggs, cruciferous veg) | MSM is part of the dietary sulfur cycle, and you also get organic sulfur from sulfur-rich foods and the amino acids cysteine and methionine in protein. | Altern Med Rev 2002 [PMID 11896744](https://pubmed.ncbi.nlm.nih.gov/11896744/) |
| Cow's milk | MSM (dimethyl sulfone) occurs naturally in trace amounts in cow's milk, making dairy one genuine dietary source of this organic-sulfur compound. | J Dairy Sci 2013 [PMID 24035021](https://pubmed.ncbi.nlm.nih.gov/24035021/) |
| Coffee, tomatoes & other plant foods | Dimethyl sulfone (MSM) is detectable as a natural trace component of milk and other foods and beverages, confirming everyday foods as real low-dose sources. | Metabolites 2018 [PMID 29642378](https://pubmed.ncbi.nlm.nih.gov/29642378/) |
