## What it is

Methylsulfonylmethane (MSM), also known as dimethyl sulfone or methyl sulfone, is a naturally occurring organosulfur compound found in small amounts in foods such as fruits, vegetables, grains, meat and dairy, and manufactured synthetically for dietary supplements. It provides a bioavailable source of sulfur and is most commonly used as an anti-inflammatory agent. Reported mechanisms include reduction of oxidative stress, down-regulation of inflammatory cytokines via NF-kB signaling, and support of glutathione status and antioxidant capacity. OptiMSM is a distilled, GRAS-designated branded grade of MSM that has been used as the active ingredient in several human clinical trials.

## Its role in longevity

MSM has been studied chiefly for knee osteoarthritis, where small, short-term randomized trials report modest improvements in pain, function or knee-related quality of life, though authors describe the benefits as small and of uncertain clinical significance, and some trials (prevention in military trainees, and a 1,500 mg/day combination for hand osteoarthritis) found no benefit over placebo. Additional human trials suggest MSM may attenuate exercise-induced muscle and joint soreness and oxidative-stress markers, and one placebo-controlled trial reported improved expert-graded signs of skin aging. Reviews and dedicated safety analyses find MSM well tolerated at up to about 4-6 g/day. Overall the evidence is limited by small samples, short durations and frequent industry funding, and this product's 1,500 mg dose sits at the low end of the 2-6 g/day range used in most positive trials. This evidence describes the compound methylsulfonylmethane and its mechanisms as studied in the literature, not this specific commercial product.

- **What the grade means:** Curated grade based on the human and mechanistic evidence for this compound.
- **Dose used in studies:** Knee-osteoarthritis trials used MSM about 1.5–6 g/day for 12 weeks; the most-cited trial (Kim 2006) used 3 g twice daily (6 g/day), and exercise-recovery studies used roughly 1.5–3 g/day.
- **Dose from food:** MSM occurs in food only at trace levels (milk ~2–6 ppm, coffee ~1.6 ppm), so dietary intake is a few milligrams a day — roughly 1,000× below the multi-gram studied doses.
- **What this grade does not say:** Weak-to-moderate: small, short, mostly industry-funded trials report modest knee-symptom benefits, and one well-run prevention trial was negative. Safety is a genuine strength (GRAS; well tolerated up to ~6 g/day). Claims beyond joint comfort, exercise recovery and skin are not well supported.
