## Methods

A two-sample Mendelian randomization (MR) study was conducted using genetic data from the MRC-IEU consortium for 461,384 individuals. Five genetic variants were identified as instrumental variables for genetically predicted glucosamine. Genetic associations of these variants with parental longevity (combined parental age at death, mother's age at death, father's age at death) were obtained. The inverse-variance weighted method was used to estimate the effect of a 1-standard deviation (SD) increase in genetically predicted glucosamine on parental longevity.

## Results

A 1-SD increase in genetically predicted glucosamine was associated with higher odds of combined parental age at death (odds ratio, 2.64) and maternal age at death (odds ratio, 1.73). No association was found with paternal age at death (odds ratio, 1.32). Sensitivity analyses did not find evidence of pleiotropic effects.

## Conclusion

Lifelong higher levels of glucosamine may increase life expectancy, with positive effects specifically associated with maternal age at death. The sex-specific findings require further investigation.
