## The question

What is the optimal dose of NMN supplementation to improve blood NAD concentration and related health markers in adults aged 40-65?

## Hypothesis

NMN supplementation will increase blood NAD concentration in a dose-dependent manner, and this increase will be associated with improvements in health markers.

## Methods

This was a post-hoc analysis of a double-blinded clinical trial with 80 healthy adults aged 40-65. Participants received a placebo or daily doses of 300 mg, 600 mg, or 900 mg NMN for 60 days. Blood NAD concentration, blood biological age, HOMA-IR, 6-minute walk test, and SF-36 survey were measured at baseline and after supplementation.

## Results

NMN supplementation resulted in a significant dose-dependent increase in NAD concentration, with a large coefficient of variation (29.2-113.3%) within groups. The increase in NAD was associated with improvements in the 6-minute walk test distance and the SF-36 score. The median effective dose of NAD for the 6-minute walk test was 15.7 nmol/L and for the SF-36 score was 13.5 nmol/L.

## Conclusion

NMN supplementation increases NAD concentration in a dose-dependent manner, which is associated with improved walking distance and quality of life. Due to high interindividual variability, monitoring NAD concentration is important for personalized NMN dosing.

## Limitations

The study noted a high interindividual variability in NAD concentration changes after NMN supplementation, indicating that a single optimal dose may not apply to everyone.
