## Methods

A panel of 10 flavonoid polyphenols was screened for senolytic activity using senescent murine and human fibroblasts. The most potent flavonoid, fisetin, was then tested in mice modeling a progeroid syndrome and in aged wild-type mice to assess its effects on senescence markers, age-related histopathology, disease markers, health span, and lifespan. Human adipose tissue explants were also used to evaluate the translatability of the results.

## Results

Fisetin was identified as the most potent senolytic among the 10 flavonoids tested. Acute or intermittent treatment with fisetin reduced senescence markers in multiple tissues in progeroid and old mice, indicating a 'hit-and-run' senolytic mechanism. Fisetin also reduced senescence in specific cell types within murine and human adipose tissue. In wild-type mice, late-life administration of fisetin restored tissue homeostasis, reduced age-related pathology, and extended both median and maximum lifespan.

## Conclusion

The natural product fisetin possesses senotherapeutic activity in mice and human tissues. Intervention with fisetin late in life was sufficient to provide significant health benefits, suggesting its potential for translation into human clinical studies.

## Limitations

The study primarily used mouse models and human tissue explants, which may not fully reflect the complexity of human aging and disease. The specific mechanisms of fisetin's cell-type specificity were not fully elucidated.
