In humans, dietary spermidine levels were assessed via food questionnaires and correlated with blood pressure and cardiovascular disease incidence.

## Results

Oral spermidine supplementation extended the lifespan of mice, reduced cardiac hypertrophy, and preserved diastolic function in old mice. It enhanced cardiac autophagy, mitophagy, mitochondrial respiration, and improved cardiomyocyte mechano-elastical properties, coinciding with increased titin phosphorylation and suppressed subclinical inflammation. Cardioprotection was not observed in mice lacking Atg5 in cardiomyocytes. In Dahl salt-sensitive rats, spermidine feeding reduced systemic blood pressure, increased titin phosphorylation, and prevented cardiac hypertrophy and diastolic dysfunction, delaying heart failure progression. In humans, high dietary spermidine levels correlated with reduced blood pressure and a lower incidence of cardiovascular disease.

## Conclusion

Oral spermidine supplementation extends lifespan and provides cardioprotection in mice and rats, likely through mechanisms involving autophagy and titin phosphorylation. Higher dietary spermidine intake in humans is associated with better cardiovascular health.
