The Smallest Amino Acid Has the Longest Resume

Glycine is a building block for collagen, glutathione, creatine and heme, and small trials show 3 grams before bed improves subjective sleep — but the studies are tiny, some are industry-affiliated, and the blockbuster aging data belong to a combination, not glycine alone.

Itemize the working life of the smallest amino acid there is. One: it makes up roughly one-third of the collagen molecule — every third position, the scaffold of skin, tendon and bone. Two: it is a required substrate for glutathione, the cell's master antioxidant. Three: substrate for creatine. Four: for heme, the iron-carrying heart of hemoglobin. Five: it feeds one-carbon methylation metabolism through the enzyme glycine N-methyltransferase. Six: in the spinal cord it is an inhibitory neurotransmitter at its own receptors. Seven: at NMDA-type glutamate receptors it is a co-agonist, a necessary hand on a switch. Eight — and this is the item that sells the powder — taken orally before bed, about 3 grams, it lowers core body temperature through peripheral vasodilation, the same thermal dip the body uses to fall asleep.

That is a lot of jobs for a molecule with two carbon atoms. The question the evidence must answer is which of those jobs supplementation actually improves.

The most defensible answer: sleep quality, modestly, in small trials; glutathione status, powerfully, but only in combination with a cysteine donor; collagen outcomes, inconsistently. Nothing yet at the scale the resume implies.

The bedtime dossier

The sleep data are the most human-facing. Small trials report that about 3 g of glycine before bedtime improves subjective sleep quality and reduces next-day fatigue and sleepiness after partial sleep restriction. A 2012 paper in the Journal of Pharmacological Sciences framed it as a new therapeutic strategy in amino acid medicine. The mechanism is unusually well characterized for a supplement: a 2014 Neuropsychopharmacology study showed the sleep-promoting and hypothermic effects are mediated by NMDA receptors in the suprachiasmatic nucleus — the brain's master clock. Glycine, in other words, nudges sleep through thermoregulation, not sedation, which fits the trial finding of less next-day sleepiness rather than more. The caveats are equally clear: the studies are small, and several were industry-affiliated.

The glutathione chapter — read the byline

The most dramatic findings carry a compound's name, not glycine's alone. GlyNAC — glycine combined with cysteine or N-acetylcysteine — has restored glutathione synthesis in older adults and, in placebo-controlled trials, improved oxidative stress, mitochondrial function, inflammation, insulin resistance, muscle strength and other aging hallmarks. Stunning resume. Now the fine print: those trials enrolled about two dozen participants, come from a single research group, tested the combination rather than glycine alone, and the benefits faded after supplementation stopped. Everything about that paragraph says 'promising, unreplicated, early.'

The collagen question, still open

You would think one-third of the collagen molecule would make collagen claims easy. Not so. Glycine-rich gelatin can raise collagen-synthesis markers, while at least one controlled trial found no added benefit. A 2023 European Journal of Nutrition trial did find collagen peptide supplementation before bed reduced sleep fragmentation and improved cognitive function in physically active men with sleep complaints — suggestive, but peptides are not powder, and one trial is not a literature.

How to hold the list

For a reader weighing a tub of glycine powder: the sleep use has the most direct support, the dose in the trials is about 3 grams, and the safety logic of an amino acid the body already makes is reassuring. The aging claims belong to GlyNAC, a different intervention awaiting independent replication. This evidence describes glycine and its mechanisms — and in several trials glycine combined with other compounds — not this specific commercial product. Taken as directed in the trials, glycine is about as gentle as supplementation gets; what it is not yet is well-proven.

The takeaway

About 3 g of glycine at bedtime improves subjective sleep quality in small trials, and glycine-plus-cysteine combinations show intriguing aging-related effects — but the human evidence is early, small-scale and largely unreplicated.
