The glucosamine–chondroitin pair is the most widely used joint supplement combination; this formula adds MSM (OptiMSM), an organic sulfur donor for connective tissue. Glucosamine feeds cartilage glycosaminoglycan synthesis, chondroitin helps cartilage hold water and resist load, and MSM lowers inflammatory and oxidative signaling in the joint.
In the MOVES trial, chondroitin sulfate plus glucosamine matched the prescription NSAID celecoxib for knee osteoarthritis pain over six months. A Cochrane review of 43 trials found chondroitin — alone or with glucosamine — modestly better than placebo for pain, with fewer serious adverse events, and the GAIT trial showed a benefit in the moderate-to-severe-pain subgroup. The evidence supports osteoarthritis symptom relief with an excellent safety profile, not a proven effect on lifespan.
