D-Glucosamine is an amino-sugar best known for joint support, but large population studies have linked regular use to lower mortality. Mechanistically it appears to act as a calorie-restriction mimetic: by feeding the hexosamine biosynthesis pathway it activates AMP-activated protein kinase (AMPK) and induces autophagy without actual caloric deprivation.
In the UK Biobank cohort (~495,000 adults), regular glucosamine use was associated with significantly lower all-cause and cardiovascular mortality over ~9 years (all-cause HR ~0.73; CVD HR ~0.42), with benefits most pronounced in people with poorer baseline cardiovascular health. A Mendelian-randomization analysis supported a causal, positive effect of higher glucosamine status on lifespan, and AI side-effect modeling has flagged it among the safest geroprotective candidates. The evidence is observational and genetic rather than from dedicated longevity randomized trials.
