Trans-resveratrol is a stilbenoid polyphenol found naturally in grape skins, red wine, peanuts and Japanese knotweed. It is the most studied of the dietary sirtuin-activating compounds. Liposomal encapsulation is used to address resveratrol's otherwise poor oral absorption and rapid metabolism.
Resveratrol is investigated as a caloric-restriction mimetic: a compound that engages the same NAD⁺-dependent SIRT1 signalling that mediates many benefits of dietary restriction. Evidence in mammals is mixed — some studies show metabolic and cardiovascular benefits while others find the lifespan effect modest — so it is presented here with that nuance.
Peer-reviewed studies on the active compound — citations link to PubMed.
“Caloric restriction has been shown repeatedly to prolong the lifespan in laboratory animals, with its benefits dependent on molecular targets... including the NAD-dependent deacetylase SIRT1.”
“The evidence that resveratrol acts as a caloric restriction mimetic in mammals is, on balance, limited, and the magnitude of any effect appears modest.”
“positive effects on bone density in the lumbar spine and femoral neck”
“At 3 months, the mean reduction in knee pain was -15.7 (95% confidence interval (CI), -21.1 to -10.3) in the resveratrol group and -15.2 (95% CI, -20.5 to -9.8) in the placebo group (absolute difference -0.6 [95% CI, -8.0 to 6.9]; p = 0.88).”
“Resveratrol treatment significantly reduced pain intensity and WOMAC scores, while increasing OKS scores, ROM, gait velocity, HGS, and overall SPPB performance (p < 0.05).”
“Rheumatoid arthritis (RA) is an inflammatory autoimmune disease, which is characterized by massive pain and destruction of synovial joints, leads to bone erosion, damage to cartilage, and disability.”
